Na maioria dos vertebrados o tridecapeptídeo alfa-MSH (hormônio estimulante do melanócito) é um hormônio fisiologicamente relevante na regulação da pigmentação da pele, causando escurecimento (Sawyer, et al., 1980; Jiang et al., 1995). O alfa-MSH também está envolvido em muitas outras funções, como crescimento fetal e comportamento (Castrucci, et al., 1990). No presente trabalho foi estudada a interação entre peptídeos melanotrópicos e membranas lipídicas sob as perspectivas da perturbação da fase lipídica e das modificações conformacionais dos peptídeos. Foram usados: dois tipos de vesículas (aniônica de DMPG - dimiristoil fosfatidil glicerol - e neutra de DMPC - dimiristoil fosfatidil colina) e dois peptídeos melanotrópicos (alfa-MSH e o análogo que apresenta maior atividade biológica, [Nle POT.4, D-Phe POT.7]-alfa-MSH (MSH-I). Por meio de ressonância paramagnética eletrônica (RPE) foram observadas alterações estruturais dos lipídios causadas pela interação dos peptídeos com a membrana aniônica. Todos os marcadores de spin usados, derivados do ácido esteárico, fosfolipídicos e derivado do colesterol, incorporados em vesículas de DMPG, na fase líquido-cristal, indicaram que os dois peptídeos induzem um decréscimo no movimento e/ou aumento da ordem das cadeias acilas, em todas as posições monitoradas. O efeito do análogo sobre os parâmetros dos espectros de RPE dos marcadores de spin foi mais evidente, e isto estaria indicando que penetra mais fundo na matriz lipídica. A dependência com o sal da interação peptídeo-lipídio demonstra que a atração eletrostática dos peptídeos carregados positivamente com as cabeças polares dos lipídios aniônicos foi necessária para uma efetiva associação dos peptídeos com a bicamada fosfolipídica. A necessidade da interação eletrostática também foi observada pela não-interação dos peptídeos com bicamadas neutras de DMPC. No desenvolvimento do trabalho foi calculado ) o coeficiente de partição baseado no efeito que estes peptídeos causam no espectro de RPE dos marcadores de spin intercalados na membrana. O coeficiente de partição obtido para peptídeo análogo, mais potente biologicamente, foi cerca de quatro vezes maior do que para o hormônio nativo. Para uma mesma concentração de peptídeo ligado à membrana, foi observado que o efeito do MSH-I sobre a estrutura da membrana é um pouco maior do que do alfa-MSH, o que estaria de acordo com uma possível penetração mais fundo na bicamada. Os espectros de dicroísmo circular (CD) em solução aquosa e no solvente indutor de alfa-hélice, o 2.2.2-trifluoroetanol (TFE) mostraram que os dois peptídeos têm estrutura um pouco diferentes em solução, embora apresentem mudanças conformacionais similares quando na presença de vesículas ou micelas carregadas negativamente. O maior efeito causado pelo análogo mais potente na estrutura da bicamada, quando comparado ao hormônio nativo, é discutido em termos de sua maior constante de associação e possibilidade de maior penetração na bicamada. Estes resultados estariam relacionados com a maior atividade e/ou prolongada ação do peptídeo análogo.

In most vertebrates the tridecapeptide -melanocyte stimulating hormone (-MSH) is physiologically relevant hormone regulating skin pigmentation, causing darkening (Sawyer, et al., 1980; Jiang et al., 1995). -MSH is also involved in many other biological functions, such as fetal growth and behavior (Castrucci, et all., 1990). The present work studies the interaction of -MSH and the biologically more active analog [Ne4, Dphe7]- -MSH with lipid vesicles by spin label electron spin resonance (Esr) spectroscopy and circular dichroism (CD). All spin labels used here, stearic acid, phospholipid and cholestane derivative labels, incorporated into anionic vesicles of DMPG (1,2-dimyritoyl-sn-glycero-3-phophoglycerol) in the liquid-cristiline phase, indicated that both peptides decrease the motional freedom of the acyl chains, at all monitored positions. The effect of the analog on the spin label ESR parameters was much more evident, and could indicate its farthest penetration into the lipid matrix. The salt dependence of the peptide- lipid interaction demonstrated that the electrostatic attraction of the positively charged residues of the peptides by the PG headgroups was necessary for an effective association of the peptides with phospholipid bilayers. The perturbation caused by the peptides on the lipid chain mobility was reduced as the ionic strength increased. Above 300 mM NaCl no perturbation could be detected. The electrostatic requirement was further evidenced by the absence of detectable binding of peptides to neutral lipid bilayers. Lipid partition coefficients were calculated based on the effect the peptides cause on the ESR spectra of spin labels incorporated in the membrane. For the biologically more potent peptide, the partition coefficient was found to be about 4-times greater than that of the native hormone. For the same concentration of the peptide bound to the membrane, MSH-I was found to cause a slightly greater effect on the membrane structure than -MSH, in accord with its possible deeper penetration into the bilayer. Cd spectra in aqueous solution and in the -helix inducing solvent 2.2.2-trifluoroethanol (TFE) showed that the two peptides have somewhat different structures in solution, though similar conformational changes occur in both peptides as a result of their interaction with negatively charged vesicles or micelles. The stronger effect caused by the more potent analog in the membrane structure, when compared to the native hormone, is discussed in terms of the higher peptide-lipid association constant and the possible deeper penetration into lipid bilayers. The results could be related to its greater activity and/or prolonged action of the peptide analog.
