Betaxantinas são produtos naturais com propriedades antioxidantes e anti-inflamatórias encontradas em algumas variedades de plantas e fungos. Nessa Tese de Doutorado foram semissintetizadas vinte e uma betaxantinas derivadas de todos os aminoácidos proteinogênicos, com exceção da selenocisteína, e algumas de suas propriedades físicoquímicas foram caracterizadas e comparadas entre si e com as betaxantinas-controle derivadas da dopamina, L-DOPA e pirrolidina. A elusiva betaxantina derivada da Lcisteína foi obtida pela primeira vez e apresenta um anel 2-tiazolínico. Ainda, foram isoladas e caracterizadas duas betaxantinas regioisoméricas derivadas da L-lisina. As propriedades de absorção e fluorescência, bem como a estabilidade hidrolítica de todas as betaxantinas foram determinadas. A capacidade antioxidante e o potencial redox das betaxantinas foram medidos e foram estabelecidas relações estrutura-propriedade por meio da análise de pares moleculares combinados. Porções fenol ou indol ligadas diretamente ao sistema conjugado imino-enamino de betaxantinas aumentam a capacidade antioxidante das betaxantinas que, de forma geral, é muito maior do que aquela observada para seus aminoácidos precursores e para antioxidantes padrão. A betaxantina derivada da cisteína possui uma capacidade antioxidante muito baixa, e nunca foi encontrada in vivo. Em contrapartida, essa betaxantina não-imínica é muito mais estável do que as demais betaxantinas derivadas de aminoácidos.

Betaxanthins are natural products with antioxidant and anti-inflammatory properties found in some varieties of plants and fungi. In this Doctoral Thesis, twenty-one betaxanthins derived from all proteinogenic amino acids, with the exception of selenocysteine, were semi-synthesized and some of their physicochemical properties were characterized and compared with each other and with the control betaxanthins derived from dopamine, L-DOPA and pyrrolidine. The elusive betaxanthin derived from L-cysteine was obtained for the first time and features a 2-thiazoline ring. Furthermore, two regioisomeric betaxanthins derived from L-lysine were isolated and characterized. Absorption and fluorescence properties as well as hydrolytic stability of all betaxanthins were determined. The antioxidant capacity and redox potential of betaxanthins were measured and structure-property relationships established through matched molecular pair analysis. Phenol or indole moieties linked directly to the imino-enamino conjugated system of betaxanthins increase the antioxidant capacity of betaxanthins, which, in general, is much higher than that observed for their precursor amino acids and for standard antioxidants. Cysteine-betaxanthin has a very low antioxidant capacity, and has never been found in vivo. In contrast, this non-iminic betaxanthin is much more stable than the other amino acid betaxanthins.
