A demanda crescente por alimentos e biocombustíveis tem levado ao aumento indiscriminado do uso de fertilizantes nitrogenados na agricultura, o que tem ocasionado impactos negativos em ecossistemas aquáticos e terrestres. Para manutenção dos altos níveis de produtividade, o uso de fertilizantes a base de nitrogênio (N) na cultura de cana-de-açúcar é inevitável, sendo amônio a fonte inorgânica de N preferencial para essa espécie. Em plantas, os transportadores responsáveis pelo transporte de amônio pertencem à família AMT (AMMONIUM TRANSPORTERS), no entanto, pouco se conhece sobre a funcionalidade desses transportadores. Para isso, esse trabalho teve como objetivo a caracterização funcional e molecular de transportadores de amônio membros da subfamília AMT1 de cana-de-açúcar em Arabidopsis thaliana. As sequências gênicas e promotoras dos membros da subfamília AMT1 de cana-de-açúcar foram identificadas por seleção em biblioteca de BACs (Bacterial artificial chromosome). Análises de expressão gênica realizadas em raízes de cana-de-açúcar demonstraram que os genes ScAMT1.1 e ScAMT1.3 são regulados pelo status de N e atuam na aquisição de amônio quando a disponibilidade exógena de N é limitante. Experimentos de localização da expressão tecido/órgão específico em A. thaliana utilizando os promotores de ScAMT1.1 e ScAMT1.3 fusionados aos genes repórters GUS e GFP demonstraram que os genes de cana-de-açúcar são preferencialmente expressos na região da endoderme/periciclo e vascular das células das raízes, sendo regulados espacialmente pela disponibilidade e fonte de N. A caracterização funcional dos transportadores de amônio identificados no genoma de cana-de-açúcar superexpressos no quádruplo mutante qko de arabidopsis demonstraram que essas plantas apresentaram alta sensibilidade ao metilamônio (análogo tóxico ao amônio), além de produzirem biomassa a níveis comparáveis ao genótipo selvagem ('Col-0') quando cultivadas exclusivamente em amônio como fonte única de N, indicando a funcionalidade de ScAMT1.1 e ScAMT1.3 no transporte de metilamônio e amônio. Análises de influxo de 15N-amônio em raízes de plantas transgênicas de qko superexpressando os genes ScAMT1.1 e ScAMT1.3 demonstraram a existência de um sistema de transporte de alta afinidade a amônio (High Affinity Transporter System - HATS) ativo, o qual é modulado em resposta ao status de N das plantas. Esses resultados indicam que os transportadores ScAMT1.1 e ScAMT1.3 de cana-de-açúcar são funcionais em raízes, atuando com propriedade distintas no transporte de amônio nessa gramínea regulando a aquisição de amônio de acordo com a disponibilidade de N no solo

The growing demand for food and biofuels has led to the indiscriminate growth in use of nitrogen (N) fertilizers in agriculture, which has caused negative impacts in water and land ecosystems. To maintain high yields, the use of N fertilizer in sugarcane cultivation is inevitable, and ammonium is the preferred N source of this species. In plants, transporters from the AMT (AMMONIUM TRANSPORTERS) family are responsible for ammonium acquisition, but little is known about the functionality of these transporters in sugarcane. This work aimed to functionally characterize members of the AMT1 gene subfamily of sugarcane in Arabidopsis thaliana. Gene and promoter region sequences of members of the sugarcane AMT1 subfamily were identified in a Bacterial Artificial Chromosome (BAC) library. Expression analysis in sugarcane roots demonstrated that ScAMT1.1 and ScAMT1.3 are regulated by N status and participate in ammonium uptake when exogenous N availability is limiting. The expression of the promoter region of ScAMT1.1 and ScAMT1.3 directing the expression of the reporter genes GUS and GFP demonstrated preferential expression in the endodermis/pericycle regions of arabidopsis roots, spatially regulated by availability and source of N. The functional characterization of ammonium transporters identified in the sugarcane genome over-expressed in the quadruple arabidopsis mutant qko demonstrated that the complemented plants exhibited high sensitivity to methyl-ammonium (a toxic ammonium analog), while producing biomass at comparable levels to the wild 'Col-0' genotype when cultivated exclusively in ammonium as the only N source, indicating the functionality of ScAMT1.1 and ScAMT1.3 in the transport of methyl-ammonium and ammonium. Analysis of 15N-ammonium influx in roots of complemented qko plants demonstrated the existence of an active ammonium High Affinity Transporter System (High Affinity Transporter System - HATS), which is modulated in response to the plant N status. These results indicated that the sugarcane ScAMT1.1 and ScAMT1.3 transporters are functional in roots, acting with distinct properties in the ammonium transport in this grass, regulating the ammonium acquisition according to the availability of soil N level
