A paracoccidioidomicose é uma micose granulomatosa sistêmica causada pelo fungo termodimórfico Paracoccidioides brasiliensis. Esta doença é a mais importante infecção fúngica na América Latina e um importante problema de saúde no Brasil, representando 51% das mortes por infecção fúngica. A homoserina desidrogenase é uma enzima oxidorredutase que atua na síntese de aminoácidos no metabolismo de fungos e plantas, não sendo encontrada em humanos. Tal fato tornou-a um alvo em potencial para o desenvolvimento de novos fármacos específicos para o tratamento da doença. O objetivo deste trabalho foi a superexpressão heteróloga em Escherichia coli, purificação e caracterização estrutural e funcional da homoserina desidrogenase da Paracoccidioides brasiliensis, a fim de fornecer base científica para posteriores estudos farmacológicos. A proteína homoserina desidrogenase foi expressa em vetor pET-SUMO na forma solúvel, obtendo-se uma quantidade média de 29,672 ± 6,828 mg de proteína por litro de expressão com aproximadamente 94% de pureza. A análise por espectroscopia de dicroísmo circular apresentou um perfil espectral de uma estrutura secundária predominantemente formada por hélices α, 45,5 ± 7,3 %, e com pouca contribuição de fitas β, 10,5 ± 7,0 %. As análises por espectroscopia de emissão de fluorescência mostraram que a proteína possui uma boa estabilidade na presença do agente desnaturante ureia, apresentando um Cm de 4,13 ± 0,21 M de ureia, e uma ampla faixa de pH na qual não há mudança conformacional, do pH 6 ao pH 10. Na análise por calorimetria diferencial de varredura foi possível verificar que a proteína possui uma alta estabilidade a temperatura ambiente, porém uma baixa estabilidade em altas temperaturas, sofrendo uma desnaturação irreversível, a Tm obtida foi de 58,65 ± 0,87 °C. Os estudos da atividade enzimática da homoserina desidrogenase por espectroscopia de absorção molecular no UV/Vis mostrou que a proteína possui um pH ótimo entre 9,35 e 9,50, um comportamento michaeliano, que a reação enzimática se dá por meio de um processo não cooperativo, que a enzima possui um KM de 0,266 mM e um VMax de 1,78 mM s-1 e que a mesma não é sensível a inibição por treonina com um IC50 de 184 ± 3,0 mM. Portanto, a expressão e purificação da proteína foram eficientes e as análises de caracterização estrutural mostraram que a proteína foi obtida na forma enovelada e enzimaticamente ativa, sendo bastante estável frente a diversos agentes externos.

Paracoccidioidomycosis is a systemic granulomatous mycosis caused by the thermodymorphic fungus Paracoccidioides brasiliensis. This disease is the most important fungal infection in Latin America and a major health problem in Brazil, representing 51 % of deaths due to fungal infection. Homoserine dehydrogenase is an oxidoreductase enzyme that acts on the synthesis of amino acids in the metabolism of fungi and plants, not being found in humans. This fact has made it a potential target for the development of new specific drugs for the treatment of the disease. The objective of this work was the heterologous overexpression in Escherichia coli, purification and structural and functional characterization of the homoserine dehydrogenase of Paracoccidioides brasiliensis, in order to provide scientific basis for later pharmacological studies. The homoserine dehydrogenase protein was expressed in pET-SUMO vector in soluble form, obtaining a mean amount of 29.672 ± 6.828 mg of protein per liter of expression at approximately 94 % purity. The analysis by circular dichroism spectroscopy showed a spectral profile of a secondary structure predominantly composed of α-helices, 45.5 ± 7.3 %, and little contribution of β-strands, 10.5 ± 7.0 %. Fluorescence emission spectroscopy analyses showed that the protein has a good stability in the presence of the denaturing agent urea, presenting a Cm of 4.13 ± 0.21 M urea, and a broad pH range in which there is no conformational change from pH 6 to pH 10. In the analysis by differential scanning calorimetry, it was possible to verify that the protein has a high stability at room temperature, but a low stability at high temperatures, suffering irreversible denaturation, the obtained Tm was 58.65 ± 0.87 °C. Studies of the enzymatic activity of homoserine dehydrogenase by molecular absorption spectroscopy in UV/Vis showed that the protein has an optimum pH between 9.35 and 9.50, a michaelian behavior, the enzymatic reaction is carried out by means of a non-cooperative process, the enzyme has a KM of 0.266 mM and a VMax of 1.78 mM s -1 and it is not sensitive to inhibition by threonine with an IC50 of 184 ± 3.0 mM. Therefore, protein expression and purification were efficient and the structural characterization analyses showed that the protein was obtained in the folded and enzymatically active form, being quite stable against several external agents.
