A raiva é uma doença tropical negligenciada (DTN) que acomete humanos, mamíferos silvestres e domésticos, cuja taxa de mortalidade mundial atinge aproximadamente 60.000 mortes humanas ao ano, o que a caracteriza como um sério problema de saúde pública, representando um grande impacto em populações mais vulneráveis, devido, principalmente, a fácil propagação de seu agente. O Lissavírus da raiva (RABV) sintetiza uma molécula glicoproteica, a RVGP (do inglês Rabies Lissavirus Glycoprotein), cuja porção exposta na superfície da membrana do vírus (chamada ectodomínio) tem função mediadora na adesão à célula hospedeira e na indução de anticorpos neutralizantes. Um controle efetivo frente a esta DTN pode ser alcançado quando abordagens em saúde pública são trabalhadas desenvolvendo, por exemplo, vacinas preventivas. Apesar do investimento, o controle da doença não tem sido o suficiente para conter sua distribuição. Desta forma, este projeto teve por objetivo otimizar a produção da molécula RVGP a partir do uso de sua porção antigênica, o ectodomínio, obtendo assim uma proteína heteróloga secretada. Para isso, foi utilizado um sistema de expressão de proteínas recombinantes baseado em células de inseto (Drosophila melanogaster Schneider 2 - S2), capaz de sintetizar de forma indutível esta molécula com possibilidade de desempenhar sua função biológica, contendo os epítopos antigênicos do lissavírus da raiva. A linhagem celular obtida através de transfecção com o agente Lipofectamine® 2000 foi denominada S2MTBip_RVGP-ecto e expressa o ectodomínio da glicoproteína do vírus da raiva de forma secretada em diferentes condições de indução e tempos de coleta. A proteína foi detectada por análises de Dot blotting, Western blotting e quantificada por ELISA. Os resultados mostram a presença de uma banda correspondente a massa molecular de aproximadamente 51 kDa, presentes no sobrenadante das culturas, reconhecida por anticorpos monoclonais (C75 e D1) e soro. A subclonagem da população inicial de S2MTBip_RVGP-ecto permitiram a seleção de uma subpopulação recombinante (S2MTBip_RVGP-ectoG5), capaz de produzir mais rRVGP-ecto, sugerindo que os valores obtidos da proteína recombinante ainda podem ser melhorados.

Rabies is a neglected tropical disease (NTD) that affects humans, wild and domestic mammals, whose mortality rate reaches approximately 60,000 human deaths each year. The rabies characterizes it as a serious public health problem, representing a major impact on vulnerable populations, mainly due to the easy spread of its agent. Rabies Lissavirus (RV) synthesizes a glycoprotein, the RVGP (Rabies Lissavirus Glycoprotein), whose exposed portion on the surface of the virus membrane (called ectodomain) has mediating function in the adhesion to the host cell and in the induction of neutralizing antibodies. Effective control of this NTD can be achieved when public health approaches are assured by developing, for example, preventive vaccines. Despite the investment, the control of the disease has not been enough to contain its distribution. Thus, this project aimed to optimize the production of the RVGP molecule from the use of its antigenic portion, the ectodomain, obtaining a heterologous protein of soluble character. For this, we used a recombinant protein expression system based on insect cells (Drosophila melanogaster Schneider 2 - S2), able to synthesize this molecule with the possibility of performing its biological function, containing the antigenic epitopes of rabies lissavirus. The cell population obtained by transfection with the agent Lipofectamine® 2000 was named S2MTBip_RVGP-ecto and expresse the ectodomain of the rabies virus glycoprotein in soluble form under different induction conditions and sampling times. Protein was detected by Dot blotting, Western blotting and quantified by ELISA. The results revealed the presence of a molecular weight band of approximately 51 kDa, recognized by monoclonal antibodies (C75 and D1) in culture supernatant. The subpopulation selection increased the rRVGP-ecto expression in relation to the population, suggesting the possibility of optimization of the production.
