A biodiversidade brasileira é vasta e promissora, rica em frutas com potencial econômico. Estas, possuem compostos bioativos, como compostos fenólicos que apresentam capacidade antioxidante e têm a propriedade de inibir enzimas chaves para a degradação de carboidratos, tais como a α-amilase e a α-glicosidase. Isso as tornam importantes agentes de redução do risco de desenvolvimento de doenças crônicas não transmissíveis. O objetivo deste trabalho foi caracterizar os compostos fenólicos e potencial atividade biológica in vitro de duas frutas nativas brasileiras: a cereja-do-rio grande (Eugenia involucrata DC.) e a uvaia (Eugenia pyriformis). Os frutos foram coletados em 2018 em diferentes regiões dos estados de São Paulo e Minas Gerais. Foram analisados os pesos médios dos acessos (polpa + casca) e realizadas as análises de açúcares totais, ácido elágico total, antocianinas monoméricas totais (apenas para os frutos vermelhos), fenólicos totais, DPPH e ORAC, ensaios da atividade de inibição de α-amilase e α-glicosidase e identificação de flavonoides. Esta última, realizada por LC qTOF-MS/MS e quantificada por CLAE-DAD. Os estudos de ancoramento molecular permitiram sugerir um modelo de interação com os compostos fenólicos e a α-amilase. Todos os acessos possuem alto teor de polpa e elevado conteúdo de açúcares solúveis em 100 g b.u. Os acessos CE035 (Salesopólis / SP) e Uvaia Comum (Rio Claro / SP) foram as amostras que apresentaram melhor capacidade antioxidante. Os flavonoides identificados na cereja-do-rio-grande vermelha pertencem às subclasses antocianinas, flavon-3-ol e flavanol; já a cereja-do-rio-grande amarela apresentaram as subclasses flavan-3-ol e flavanol. A uvaia apresentou duas subclasses flavan-3-ol e flavanol. O ácido elágico foi detectado em maior conteúdo em 95% os acessos do que o conteúdo de compostos fenólicos. Os acessos CE057 (Ibitinga / SP) e UV068 (Cabo Verde / MG) foram as amostras mais eficientes em inibir a α-amilase. O extrato de uvaia que mais inibiu α-glicosidase foi a de Uvaia Pera (Rio Claro / SP). Diante dos resultados de score do ancoramento molecular, todos os compostos fenólicos obtiveram média de ranqueamento mais significativa do que a acarbose (inibidor de α-amilase), exceto o dímero de proantocianidina. Desta forma, conclui-se que frutos cereja-do-rio-grande e 8 uvaia possuem boa capacidade antioxidante e podem ser considerados bons inibidores de α-amilase e α-glicosidase, os resultados de ancoramento molecular validaram os resultados visualizados nas análises in silico. A proposta é que a sociedade se aproprie desses resultados para obter retorno com o uso e valorização adequados do seu patrimônio genético.

The Brazilian biodiversity is vast and promising, rich in fruits with economic potential. These fruits have bioactive compounds, such as phenolic compounds that have antioxidant capacity and have the property of inhibiting key enzymes for carbohydrate degradation, such as α-amylase and α-glucosidase. This makes them important agents for reducing the risk of developing non-communicable chronic diseases. The aim of this work was to characterize the phenolic compounds and potential in vitro biological activity of two Brazilian native fruits: cereja-do-rio-grande (Eugenia involucrata DC.) and uvaia (Eugenia pyriformis). The fruits were collected in 2018 in different regions of the states of São Paulo and Minas Gerais. The average weights of the accessions (pulp + peel) were analyzed and the analyses of total sugars, total ellagic acid, total monomeric anthocyanins (only for red fruits), total phenolics, DPPH and ORAC, α-amylase and α glucosidase inhibition activity assays, and flavonoid identification were performed. The latter, performed by LC-qTOF-MS/MS and quantified by CLAE-DAD. Molecular docking studies allowed suggesting an interaction model with phenolic compounds and α-amylase. All accessions have high pulp content and high content of soluble sugars in 100 g w.b. The accessions CE035 (Salesopólis / SP) and Uvaia Comum (Rio Claro / SP) were the samples that showed the best antioxidant capacity. The flavonoids identified in the red cereja-do-rio-grande belong to the anthocyanins, flavon-3-ol and flavanol subclasses, while the yellow cereja-do-rio-grande showed the subclasses flavan-3-ol and flavanol. Uvaia presented two subclasses, flavan-3-ol and flavanol. Ellagic acid was detected in higher content in 95% of the accessions than the content of phenolic compounds. The accessions CE057 (Ibitinga / SP) and UV068 (Cabo Verde / MG) were the most efficient samples in inhibiting α-amylase. The uvaia extract that most inhibited α-glucosidase was that of Uvaia Pera (Rio Claro / SP). Given the molecular docking score results, all phenolic compounds obtained more significant rank average than acarbose (α-amylase inhibitor), except proanthocyanidin dimer. Thus, it is concluded that cereja-do-rio-grande and uvaia have good antioxidant capacity and can be considered good inhibitors of α-amylase and α-glucosidase, the molecular docking 10 results validated the results visualized in the in silico analyses. The proposal is that society appropriates these results to obtain return with the proper use and valuation of its genetic heritage.
