Com o objetivo de estudar as relações quantitativas entre a estrutura química e a atividade antimicrobiana de análogos à nifuroxazida (5-nitro2-furfurilideno 4-hidroxi benzidrazida), prepararam-se quatorze 5-nitro-2-furfurilideno benzidrazidas X3,X4,X5-substituídas em que X3 e X5 = H e X4 = NO2, Br, Cl, H, CH3, OCH3, OH, NH2 COCH3, OC2H5, CF3, N(CH3)2, SO2NH2 e X3,X4,X5 = OCH3. Nove entre os quatorze compostos obtidos ainda não estão descritos na literatura. Os compostos obtidos foram identificados e/ou caracterizados por seus espectros de I.V., RMN1H e RMN13C, e seus graus de pureza determinados pelas respectivas análises elementares de C, H e N e pelos intervalos de fusão. Determinaram-se os valores de freqüência de absorção do grupo carbonila na região do I.V., em DMSO, como medida de sua polaridade. Este parâmetro foi utilizado como medida do efeito eletrônico dos grupos substituintes estudados. Determinou-se também o coeficiente de partição de cinco compostos através do método de Shake-flask, utilizando-se, para partição, o sistema l-octanol/tampão fosfato, obtendo-se excelentes correlações entre os valores experimentais e calculados. Determinou-se a concentração mínima inibitória, MIC, dos compostos obtidos frente a cepa ATCC 25923 de S.aureus, utilizando-se o Logl/Mol como medida da potência antimicrobiana. O estudo das relações quantitativas entre a estrutura química e a atividade antimicrobiana, QSAR, foi realizada através da aplicação do método stepwise aos valores de Logl/Mol em função de parâmetros estruturais, experimentais e de literatura, relacionados com os efeitos hidrofóbico, eletrônico e de polarizabilidade dos grupos substituintes.

Aiming the study of quantitative relationships between chemical structure and antimicrobial activity of nifuroxazide (5-nitro-2-furfurylidene 4-hydroxybenzidrazide) analogues, fourteen X3,X4,X5-substituted 5-nitro-2-furfurylidene benzidrazides were prepared, where X3 and X5 = H and X = NO2, Br, Cl, H, CH3, OCH3, OH, NH2 COCH3, OC2H5, CF3, N(CH3)2, SO2NH2 and X3,X4,X5 = OCH3. Nine of these compounds have not been described in the literature yet. The compounds obtained were identified ou characterized through their IR, lH NMR and 13C NMR spectra. Their purity was determined through C, H and N elemental analysis, and melting interval. Values of carbonyl absorption frequency in the IR region were determined in DMSO, and taken as measure of polarity. This parameter was used as measure of the electronic effect of the studied substituent groups. The partition coefficient of five compounds was determined using the Shake-fIask method and the l-octanol/phosphate buffer system. Excellent correlations between experimental and calculated values were obtained. The minimal inhibitory concentration, MIC, of the compounds was determined against S. aureus, ATCC 25923 strain. Logl/Mol was used as measure of antimicrobial potency. The study of the quantitative structure-activity relationships, QSAR, was carried using the stepwise regression of Logl/Mol values as function of experimental and theoretical parameters related to the hydrophobic, electronic and polarizability effect of substituents.
