O congelamento do suco de folhas de plantas de fumo infetadas pelo VAP, a aplicação de centrifugações diferenciais e dois gradientes de densidade de sacarose permitiram purificar o vírus e separar perfeitamente as partículas pequenas (55 mu) das partículas grandes (200 mu). Essa técnica apresentou resultados mais satisfatórios do que outras descritas na literatura. Os testes de infetividade, feitos com as partículas separadas, mostraram que somente as partículas grandes (200 mu) são infetivas. A separação das partículas permitiu o preparo de antissoros específicos. Os testes serológicos cruzados, os testes de absorção cruzada e as observações dos precipitados serológicos ao microscópio electrónico, constituem provas convincentes de que as partículas pequenas (55 mu) e partículas grandes (200 mu) do VAP são serológicamente idênticas. Os tratamentos térmicos induziram sensíveis modificações na morfologia das partículas do VAP. Inicialmente houve uma fragmentação em partículas menores. Estas, em seguida, romperam-se em pequenos anéis e finalmente se transformaram numa massa amorfa constituída, provavelmente, de grupos de subunidades proteicas. Essas modificações puderam ser interpretadas, principalmente, pelos resultados obtidos nos testes serológicos de dupla difusão em ágar e pelas observações ao microscópio eletrônico. O VAP apresentou relações serológicas muito distantes quando comparado com alguns isolados do grupo do TRV. Com base nos resultados obtidos pode-se considerar o VAP como pertencente a um Serotipo diferente dos demais vírus do grupo do TRV. O diagnóstico serológico da presença do VAP no suco (não tratado) de folhas de fumo, pimentão e tomate foi negativo. Quando, porém, o vírus contido no suco foi submetido a uma certa concentração e purificação os resultados foram sempre positivos.

Juice from tobacco plants infected with VAP, stored at -20°C up to four months, thawed, centrifuged at alternate cycles of low and high speed plus two cycles of sucrose density gradient, permitted not only the purification, but also an almost perfect separation of the short and long particles (55 mu and 200 mu respectively). The results were better than those obtained with other purification techniques previously described. Infectivity tests performed with separated particles indicate that only the long particles (200 mu) are infective. The separation of the particles allowed the preparation of specific antisera. Cross serological tests, absorption tests and observations of the serological precipitates under the electron microscope have shown that short and long particles are serologically identical. Heat treatment induced changes in the particle morphology. At first there was a fragmentation in small particles, followed immediately by a rupture of particle these in small rings and finally reaching the size of protein subunit groups. These modifications could be interpreted specially by serological double diffusion tests in agar and by the observations of the heat treated supernatants under the electron microscope. The results of the cross serological tests between the VAP and several strains of the TRV-group have shown that the former presented only distant serological relationship with one strain of TRV from Scotland and with two strains of TRV from Holland. It seems reasonable to consider the VAP as belonging to a completely different Serotype-group within the TRV-group. Serological diagnosis of the VAP in crude sap preparations from tobacco, sweet pepper and tomato has not been possible yet. When the virus from the sap was partially concentrated the serological diagnosis became possible without any difficulty.
