Com o objetivo de enriquecer culturas de bactérias metanogênicas, verificar as morfologias predominantes e avaliar seu potencial de degradar pentaclorofenol (PCP), estudou-se um consórcio metanogênico que já havia sido inibido quando exposto a concentração de 10 mg/L de PCP. Foram realizados ensaios com 5mg PCP/g SSV utilizando-se reatores de 250 e 1000mL. Tais ensaios foram denominados ensaio 1 e 2, respectivamente. Ácidos acético, propiônico, butírico e o álcool metanol foram adicionados como fontes de carbono. Para verificação do potencial de biodegradação foram monitorados PCP. TCP, DCP e o gás CH4. As taxas de remoção de PCP, no ensaio 1, variaram entre 75 e 97% e, no ensaio 2, entre 90 e 97%. Os consórcios metanogênicos produziram cerca de 0,07 moles/L e de 0,03 moles/L de CH4, respectivamente. Os principais tipos bacterianos metanogênicos encontrados foram Methanosarcina sp, Methanococcus sp., Methanothrix sp., Methanobacterium sp., Methanobrevibacter sp. e, possivelmente, Methanosphaera sp. Bactérias não-metanogênicas na forma de cocos, espirilos e bacilos também estavam presentes. Não houve predominância de tipos morfológicos metanogênicos ou não, que pudessem ser relacionaods como possíveis degradadores de PCPentre 75 e 97% e, no ensaio 2, entre 90 e 97%. Os consórcios metanogênicos produziram cerca de 0,07 moles/L e de 0,03 moles/L de 'CH IND.4', respectivamente. Os principais tipos bacterianos metanogênicos encontrados foram Methanosarcina sp, Methanococcus sp., Methanothrix sp., Methanobacterium sp., Methanobrevibacter sp. e, possivelmente, Methanosphaera sp. Bactérias não-metanogênicas na forma de cocos, espirilos e bacilos também estavam presentes. Não houve predominância de tipos morfológicos metanogênicos ou não, que pudessem ser relacionados como possíveis degradadores de PCP

This work reports on experiments aiming at the evaluation o f the potential for pentachlorophenol degradation by methanogenic consortium. The inoculum used was previously inhibited to PCP at concentrations o f 1 O mg!L. The experiments were carried out using reactors with total volumes of 250 mL ( experiment 1) and 1000 mL (experiment 2) and substrate containing 5mg PCP/g volatile suspended solids. Acetic, propionic and butiric acids besides methanol were added as carbon sources. The concentration o f pentachlorophenol, trichlorophenol, dichlorophenol and methane gas were monitored in order to verify the potential of degradation. The PCP removal rates ranged from 7 5 and 97% in the experiment 1 and from 90 to 97% in the experiment 2. The methane concentrations 0.07 mol/L and 0.03 mol/L, respectively. Methanosarcina sp., Methanococcus sp., Methanothrix sp., Methanobacterium sp., Methanobrevibacter sp. were the main methanogenic bacterias observed in the reactor. Another genus, probably Methanosphaera sp., was also observed. Non- methanogenic bacteria as cocci, spirilla and rods were also observed . It was not observed none particular cell morphology that could be related with PCP degraders or co-metabolism activity, besides the fluorescent methanogenic sarcinas predominance.
